Some may say Dead Space 2, but I would probably say BOTH.
You can get ME2 at a steal if you look hard enough (some may have it under 20 new) and if you find Dead Space 2 at any kind of discount (online tends to have better deals), then you might get away with both of them for the price of one.
But if you're looking to get just one, I still say Mass Effect 2 (then wait for Dead Space 2 to lower in price) cause it's easier to find it cheaper than retail.

Personally, i'd get Mass Effect 2. Although, a better alternative might be to just rent both. Mass Effect 2 is just a big campaign, so once you're done, there's no multiplayer. You can replay it, but i haven't. Dead Space 2 has a really good campaign, from what i've heard. And i also heard the multiplayer was lack luster. So renting both might be a good idea.
But if you want to buy one of the two, it just depends on what you want. Do you want a really deep RPG which is a lot of fun? Go with Mass Effect 2.
Do you like scary games that are a bunch of fun to play? And maybe a little multiplayer? Go with Dead Space 2.

Mass Effect 2 can be had for 16 bucks right now for the PC at Amazon, thats for the digital download copy too. if you have the money, I would snag deals on both of them, I haven't played M.E.2 yet (don't kill me!.) but have gone through Dead Space 2. and it's a great game.

If your not playing online Mass Effect 2 will last longer than Dead Space 2. The singleplayer for Dead Space 2 is pretty scary, but shorter. So I would say it depends more on what you are going to do in the long run: play online (Dead Space 2) or play singleplayer (Mass Effect 2).
