A Big Daddy guards the Little Sisters, and the Little Sister recycle Rapture's biological energy source, ADAM. ADAM can be used to modify a user's body in certain ways, so in the game it is advantageous to kill a Big Daddy and get ADAM from the Little Sister.

Well, for the main game, while the Experimental MIRV is the strongest weapon, it's low on ammo so I'd say Lincoln's Repeater, in the Museum of History (I think) Also, if you have the DLC's, then check out these 2 that were invaluable- In Operation Anchorage, after the end of the simulation, the Gauss Rifle is in the reward vault. It's an energetic, supercharged, hunting rifle with a scope. In Broken Steel, during the assault on Adams AirForce base, you are given a Tesla Cannon, which is a energy weapon, like a rocket launcher. Super accurate and massively powerful.
